Kom was born in the Indian state of Manipur’s rural Kangathei village, Moirang Lamkhai. Kom attended the Moirang Loktak Christian Model High School. She had grown quite interested in running the 400 meters and throwing the javelin. Dingko Singh, a fellow Manipuri, had won the gold medal at the Asian Games in Bangkok in 1998. Like others, she was motivated and considered giving boxing a shot. In Imphal, she began her training under the direction of her first coach, K. Kosana Meitei. She then received instruction from Imphal-based Manipur State Boxing Coach M. Narjit Singh.

In 2005, Kom wed football player Karung Onkholer. They have three sons collectively. Kom is a proponent of animal rights and a member of PETA India. Harper Collins released her autobiography in late 2013. In 2014, the biographical film Mary Kom, starring Priyanka Chopra and based on the life of Mary Kom, was released. The Indian president proposed her for election to the Rajya Sabha in 2016.
